Modeling the survival of <i>Campylobacter jejuni</i> in raw milk considering the viable but non‐culturable cells (<scp>VBNC</scp>)

چکیده
Campylobacter spp. cannot grow in raw milk, but it is able to transform into a viable non-culturable (VBNC) state enabling the survival such harsh conditions. In this study, jejuni milk was investigated taken consideration colony-forming units (CFUs) and VBNC cells. CFU from two different strains of C. (DSM 4688 BfR-CA-18043) were enumerated at three temperatures (5°C, 8°C, 12°C). parallel, viability real-time PCR conducted quantify intact putatively infectious (IPIUs) (comprising bacteria). The data generated used model during storage. Here, one-step fitting approach performed using parameter estimates an intermediate two-step fit as starting values generate tertiary models. Different primary equations (Trilinear Weibull) required IPIU data. Strain-specific linear secondary models analyze effect storage temperature on maximum specific inactivation rate time first decimal reduction could be modeled by strain-independent model. developed for differ significantly their predictions, example, one log10 reduction. Taken that revert culturable storage, our results underline importance considering not only avoid underestimation milk.

The viable but nonculturable state in bacteria.
It had long been assumed that a bacterial cell was dead when it was no longer able to grow on routine culture media. We now know that this assumption is simplistic, and that there are many situations where a cell loses culturability but remains viable and potentially able to regrow. This mini-review defines what the "viable but nonculturable" (VBNC) state is, and illustrates the methods that ca...
متن کاملPhysiological characterization of viable-but-nonculturable Campylobacter jejuni cells.
Campylobacter jejuni is a pathogenic, microaerophilic, gram-negative, mesophilic bacterium. Three strains isolated from humans with enteric campylobacteriosis were able to survive at high population levels (10(7) cells ml-1) as viable-but-nonculturable (VBNC) forms in microcosm water. The VBNC forms of the three C. jejuni strains were enumerated and characterized by using 5-cyano-2,3-ditolyl te...
متن کاملResuscitation of Vibrio vulnificus from the Viable but Nonculturable State.
Like many other gram-negative bacteria, the human pathogen Vibrio vulnificus is induced into a viable but nonculturable (VBNC) state by incubation at low temperatures. The ability of any bacterium to resuscitate from this dormant state would appear to be essential if the VBNC state is truly a survival strategy. The question as to whether the culturable cells which appear following removal of th...
متن کاملThe viable but nonculturable state and cellular resuscitation
Aquatic microbial ecologists have long recognized that portions of bacterial populations seem to disappear from natural water bodies during certain times of the year, only to reappear at other times. In the case of V. vulnificus, numerous studies have documented the difficulty researchers experience in culturing this bacterium during cold months, presumably due to “die-off” of the population. T...
